钙黄绿素活性染料选择指南

 钙黄绿素 AM 是一种膜渗透性荧光酯酶底物,通常用于检测细胞活性和质膜完整性。作为一种近中性的底物,钙黄绿素 AM 可以自由、无创地扩散穿过质膜并渗透到活细胞的胞质溶胶中。一旦进入细胞内,代谢活跃细胞中的胞质酯酶会迅速将非荧光钙黄绿素 AM 底物水解成钙黄绿素产物,并由具有完整质膜的细胞保留。由于死细胞缺乏酯酶活性,因此仅标记活细胞。在蓝光激发下,钙黄绿素发出强烈的绿色荧光,与细胞酯酶的活性和样品中存在的活细胞数量成正比。与其他活细胞标记试剂(如 BCECF AM 或羧基荧光素二乙酸酯)相比,钙黄绿素 AM 荧光更亮、光稳定性更好,低细胞毒性有利于细胞追踪、细胞凋亡和细胞毒性的研究。

In live cells, non-fluorescent calcein AM is converted to a fluorescent calcein product after AM ester hydrolysis by intracellular esterases.

Readout

Live cells are highly fluorescent and easily distinguishable from dead cells, which are non-fluorescent.
